Hi everyone,
I'm riding a 2012 CAAD10 Ultegra which has 6700 Shimano group set on it. Recently there's been a loud clicking noise coming from the rear derailer with every rotation of the crank. After closer inspection it appears the derailer is lifting at a certain point when the chain runs through it. I've played around with the tension on the back cable running into the derailer and that fixes it for a short period of time, but the noise once again returns when I next ride. I'm wondering if others have come across this issue before? I'm hoping it's either the cable or the chain... I don't really want to spring for a new casette or worse a new rear derailer!

Is it possible you have bent the outer plates slightly on a chain link by dropping the chain off the chain rings or similar? Is there a stiff link at one point on the chain? New chain if so.
Also possible the derailleur hanger is bent. I recently had a problem with my rear ultegra 6700 dreailleur and have taken it off. The derailleur hanger broke some time ago and the derailleur dropped into the spokes locking the rear wheel. Despite replacing the hanger and the shifter cable it has never been spot on in the middle of the cassette. Turns out the cage that holds the jockey wheels must have been bent when the hanger broke and I hadn't picked it up.
I can confirm 6600 ultegra doesn't have the range of later 6700, I can't run the 30t cassette with the 6600 derailleur that is temporarily on there.

did you recently change cables? check rear derail adjustment again- could be the chain trying to "grab" the next gear up if youve had cable stretch. if youre not sure how to tune the rear deraileur get a friend who does or watch some youtube vids. the settings are pretty sensitive (about 1/4 turn of the adjuster on the rear deraileur barrel can cause clicking noise)

This reminds me of a problem where there was a regular noise and the chain would sometimes skip in the highest gear. I found it was all due to a stiff chain link.
If it's a regular pattern, try to work out which component is exactly in synch with it. Is the crank position always exactly in time with the noise. You could paint a marker on one chain link to see if that link position is in time with the noise. Do the same with the cassette then investigate whatever component is in synch with the noise.

Are you sure its the derailleur? Recently I heard a regular click from my left crank side and thought "its the BB" so got a new BB and it turned out to be the pedal.

Since a typical chain is 110-114 links long if it was a stiff link it would be occurring every two - three turns, depending on the chainring used.
Is this whilst pedalling backwards or forwards?
I don't understand the description of the derailer 'lifting'.
