Yes, the lever must be on the left or right side of the fork.










It may also point forward or back, up or down.
(but some skewers with long handles may work better with the lever on the opposite side of the disc rotor, "some" reports of rotor snag which could be possible and would be nasty)

Don't "cross" the lever over the fork blade; you can't get your fingers behind it when you want to undo it.

Rear lever on the left side so it doesn't foul the derailleur. Front also on the left because it looks untidy when they're on opposite sides.
Skewers hooking rotors has only been reported with *some* skewers, and only when incorrectly tightened with the lever left in the open position.

Your choice.
I like to have the leaver when closed to have free room at the rear so I can get my hand to it. I don't like the leaver to get dirty ... so I resist having it on the rear drive side. Some like to consider 'what if a stick comes up and hits it' .. more of a MTB thing.
But suit yourself.

If you have a support van you will also have a mechanic .. and they will set it up to suit themselves.

Oh I just corrected my post above. The rear lever is never on the drive side. The derailleur is in the way, and the cable.
So the rear is on the left, and the front is on the left.

I have a through-axle with the front lever on the right which irritates a little. Maybe I should flip the fork around

The rear made sense already. So vanity and wanting to look pro determines which side of the forks. Cool.....bike may as well look pro for the amount of money I've spent on this lifestyle

as Duck said, it leaves a gap behind the clamp to get your fingers onto. Not as critical will skinny steel forks, but makes a difference with big fat ones. Means you can use this technique for closing too:
